Comment/Description

Topic:

Density gradient centrifugation is widely used for separating cellular organelles or macromolecular complexes. During the course the biophysical background of ultracentrifugation will be discussed as well as experimental design (e.g. centrifugation speed, rotors, chemical compounds of gradient solutions) for specific scientific goals. As an example we will perform sucrose density gradient centrifugation with mitochondrial extracts to separate and analyze the mitochondrial ribosomal subunits.

 

Schedule:

Day 1:

- Theoretical introduction into sucrose gradient ultracentrifugation and Biocomp gradient station; Discussing the experimental workflow;

- Isolation of mitochondria from cultured human cells;

- Gradient preparation;

- Loading of mitochondrial lysates on a gradient and starting the ultracentrifuge.

 

Day 2:

- Fractionating the sucrose gradient using Biocomp gradient station;

- Discussing the results.

 

At the end of the course the participants should be able to plan and perform sucrose gradient ultracentrifugation and adjust experimental parameters for their specific scientific goals.

The course is part of admission "GGNB_Methods_2020_SoSe_February_April".
You can register and sign out for courses in April from 1-20 February. The slots will be assigned around 25 February.
In exceptional circumstances, you can still sign out before 29 February.
